Yeast cells were grown in 150 ml of B medium (growth medium with no sulfur element) in the presence of a repressing amount (1 mM) of L-methionine as sulfur source. When the cells reached a density of about 107 cells per ml, they were harvested by filtration and washed with 150 ml of B medium. The cells were then suspended in 100 ml of B medium without methionine at 28°C
